J Ethnopharmacol:川芎-当归药对通过促进线粒体自噬来缓解APP/PS1小鼠的认知缺陷

探讨川芎-当归药对(CDHP)在APP/PS1小鼠体内抗阿尔茨海默病(AD)的作用机制。

背景:线粒体功能障碍与各种突触损伤有关,包括轴突传导、神经递质释放和再摄取,这可能在阿尔茨海默病(AD)的早期阶段表现出来。神经元活动显著依赖于功能性线粒体,正常神经元功能的代谢成本很高。线粒体功能障碍加剧了阿尔茨海默病的病理进展,这是阿尔茨海默病的一个关键病理环节。线粒体自噬是清除受损和功能失调的线粒体的重要机制,从而在维持线粒体功能稳态和神经元突触可塑性方面发挥着至关重要的作用。研究表明,线粒体功能障碍会导致AD患者的神经变性,而线粒体自噬增强已被证明可以减轻动物模型中tau蛋白的过度磷酸化和记忆缺陷。

结果:被动回避实验和Morris水迷宫(MWM)结果表明,CDHP显著减轻APP/PS1小鼠的认知缺陷,进一步的研究发现,CDHP、突触体素(SYN)和突触后密度蛋白95(PSD-95)可以挽救海马CA 1和CA 3区树突棘密度的显著降低。CDHP可显著降低大鼠海马神经元中Caspase-12的表达水平,增强Bcl-2/Bax的表达,从而抑制神经元凋亡。

CDHP通过减少异常线粒体和增加COXIV的表达来改善AD模型中线粒体的形态和功能。CDHP处理后APP/PS1小鼠自噬体几乎不存在,而p62和LC3B的表达增加,并且CDHP还增加APP/PS1小鼠和AD样细胞模型中FUNDC 1PGAM 5的表达。

结论:这些发现表明,CDHP减轻APP/PS1小鼠的认知功能障碍,通过增强线粒体自噬,以减少神经元损伤。该研究突出了CDHP在AD中神经保护作用的潜在机制,并为未来的治疗方法提供了一条潜在的途径。
